赭曲霉毒素A和玉米赤霉烯酮-二联胶体金免疫层析试纸条的制备及应用

【背景】真菌毒素为真菌的有毒次级代谢产物,混合污染时毒性显著增强,可对人类和动物健康造成严重伤害。制备二联胶体金免疫层析试纸条,实现对常见真菌毒素混合污染的快速监测,具有重要意义。【目的】制备赭曲霉毒素A (Ochratoxin A,OTA)和玉米赤霉烯酮(Zeralenone,ZEN)金标单克隆抗体,基于免疫层析原理,采用竞争反应模式,建立二联胶体金免疫层析检测法用于污染样品中OTA和ZEN的同时快速检测。【方法】采用柠檬酸钠还原法制备胶体金颗粒,并标记获得两种真菌毒素金标单克隆抗体,通过优化相关条件,建立稳定的二联胶体金免疫层析检测方法,用于同时检测谷物和饲料样品中的OTA和ZEN。【结果】制备的OTA和ZEN二联胶体金试纸条对OTA和ZEN的检测限分别为0.625 ng/mL和1.25 ng/mL,且与谷物和饲料中其它真菌毒素(黄曲霉毒素B1、伏马毒素B1、桔青霉毒素、展青霉毒素和呕吐毒素)均无交叉反应,人工添加试验结果准确。对天然样本检测结果表明该方法与LC-MS/MS一致性良好。【结论】本研究制备的二联胶体金试纸条可用于实际样品中OTA和ZEN的同时快速筛查。     

Newly-formed photosynthates and the respiration rate of girdled stems of Korean pine (Pinus koraiensis Sieb. et Zucc.)

A stem-girdling experiment was carried out on an evergreen conifer, the Korean pine (Pinus koraiensis Sieb. et Zucc.), in mid summer in Northeast China. A 50 % higher respiration rate at the upper part of the stem was observed 3 d after stem girdling, and a stable higher rate (1.2–2.8 times) one week later. However, no higher soluble sugar or starch contents were found in the upper bark of the girdled stems in measurements over three weeks. These findings indicate that most of the newly-formed photosynthates were consumed by the high respiratory activity; this is also implied by the strong correlation between the photosynthetic photon flux over the canopy (PPF) and respiration at the upper parts of girdled stems. Moreover, the maximum PPF and cumulative PPF one day before measurement (PPFmax-Y and CPPF-Y, respectively) were closely correlated with the respiratory difference between the upper and the lower parts, but no such correlation was found with the instantaneous PPF (PPF-I) and cumulative PPF on the current day from sunrise to measured time point (CPPF-C). This shows that photosynthates newly formed by canopy needles need at least one day for transportation in order to increase the stem respiration at tree breast height.     

Gold Nanoparticles from Induced Au3+→Au0 Reaction in Polyvinyl Alcohol Molecules in Presence of Sucrose in Hot Water

In hot water (50–60°C), polyvinyl alcohol (PVA) molecules have coordination reaction with Au³⁺ cations, forming an Au³⁺-PVA polymer complex. In the proposed model reaction in small templates, the complex converts to Au⁰ capping in PVA molecules. Adding sucrose (5–10 times the PVA in mass) in a typical batch promotes Au³⁺→Au⁰ reaction, showing absorption coefficient α in Au⁰ surface plasmon band to be enhanced as much as 28 times the value in reaction with PVA. The band shifts at 547 nm from 566 nm (α  =  21.4 cm⁻¹ mol⁻¹) in the PVA sample. Drying Au⁰-PVA/sucrose (2–5 wt% Au⁰) colloid at 60–70°C and then heating at 450°C in air burns off the organic part, leaving behind a light ash colored powder with Au⁰ nanoprisms or nanofibrils (∼30 nm average width). X-ray diffractogram has six reflections, (111), (200), (220), (311), (222), and (400), of Fm3m fcc Au⁰ of lattice parameter a  =  0.4080 nm. The powder has photoluminescence in transversal and longitudinal Au⁰ plasmon bands of 535 and 585 nm, respectively.     

GICA和CLIA联合检测SARS-CoV-2特异性抗体的检测策略研究

评价胶体金免疫层析法(GICA)与化学发光法(CLIA)联合检测对降低新型冠状病毒(SARS-CoV-2)特异性抗体假阳性的效果。收集2020年1月22日至2020年3月5日就诊于川北医学院附属医院及南充市中心医院的19例SARS-CoV-2确诊患者不同时段的血清33份,55例非SARS-CoV-2、其他病原体感染及自身免疫性疾病患者的血清55份,采用GICA和CLIA分别对血清SARS-CoV-2 IgM、IgG进行检测,并对结果进行分析。GCIA检测SARS-CoV-2 IgM、IgG的敏感性分别为100.0%、94.74%,与CLIA(92.86%和100.0%)比较没有差异(P>0.05);GCIA检测SARS-CoV-2 IgM、IgG的特异性分别为70.91%、74.55%,明显低于CLIA的特异性(98.18%和89.09%)(P<0.01);两种方法检测SARS-CoV-2 IgM、IgG结果具有一致性(P<0.001),Kappa值分别为0.434,0.406;ROC曲线分析发现,GCIA检测SARS-CoV-2 IgM、IgG的AUC分别为0.855、0.846,明显低于CLIA(0.955和0.945)(P<0.05)。两种方法联合检测SARS-CoV-2 IgM、IgG的敏感性分别为92.86%、94.74%,特异性分别为100.0%、100.0%;ROC曲线分析显示,联合检测SARS-CoV-2 IgM、IgG的AUC分别为0.964、0.974,高于两种方法的单独检测。GICA和CLIA联合检测能有效提高SARS-CoV-2 IgM和IgG的检测特异性,值得临床推广应用。

以“环境微生物学”为载体的环境生态工程专业本科生科创能力培养

环境生态工程是一个设立较晚的新专业,其基本的教学体系和专业课程设计还有待摸索和完善。"环境微生物学"作为环境生态工程专业普遍设置的专业必修课,其授课内容和形式如何适应专业整体的培养需求是一个值得探讨的问题。我们结合所在学校、学院的特点和优势,提出将融入海洋特色和生态学思想的"环境微生物学"作为载体,结合课上理论教学、学生课堂报告、课下专题培训和实验技能培养等形式,培养环境生态工程专业本科生的科创能力。这可为同专业开设"环境微生物学"课程的教师提供教学参考,也可以为其他院校环境生态工程专业本科生科创能力的培养提供借鉴。     

Transition metal complexes bearing atropisomeric saturated NHC ligands

From achiral imidazolinium salts, chiral transition metal complexes containing an N-heterocyclic carbene (NHC) ligand were prepared (metal = palladium, copper, silver, gold, rhodium). Axial chirality in these complexes results from the formation of the metal-carbene bond leading to the restriction of rotation of dissymmetric N-aryl substituents about the C–N bond. When these complexes exhibited a sufficient configurational stability, a resolution by chiral high-performance liquid chromatography (HPLC) on preparative scale enabled isolation of enantiomers with excellent enantiopurities (>99% ee) and good yields. A study of the enantiomerization barriers revealed the effect of the backbone nature as well as the type of transition metal on its values. Nevertheless, the evaluation of palladium-based complexes in asymmetric intramolecular α-arylation of amides demonstrated that the ability to induce an enantioselectivity cannot be correlated to the configurational stability of the precatalysts.     

医学研究生临床生化专题全英教学的改革与实践思考

为培养高质量国际化有才能的医学研究生,对于已经学习过生物化学的国际医学留学研究生,开展了以与临床密切相关的生物化学专题为主要教学内容的全英文授课的教学实践和课程建设,取得了很好的教学效果.临床生化专题全英教学课程也可能适用于国内生物化学和医学相关专业的研究生,在今后的教学实践中需要继续研究和探索,以提高国内医学相关研究生的专业综合素质.     

Narratives of success among Irish and African Caribbean migrants

This paper compares the narratives of two men in midlife who migrated to the UK from Ireland and from the Caribbean as children, in the middle of the last century. We examine how success is narrated over the life course to show how migrants’ positioning of themselves differs from the ways in which they are positioned by outsiders, including in policy and public discourse. We conclude that while outsider narratives often polarise success and failure, insider understandings of success are dynamic and culturally and historically situated.
